Abstract:
The objectives of this project are to survey and develop an understanding of olive gnome diversity and to initiate the development of a highly informative molecular genetic map of olive.

Utilising the marker systems which optimise detection of genome variation at inter or intraspecies level a more comprehensive study of olive diversity in the different accessions of olive germplasm (varieties, wild ecotypes, related species) will be done. This includes a comprehensive analysis of DNA content, karyotype and study of molecular marker systems which highlight variability in specific genomic elements (chloroplast DNA, nuclear low copy, cDNA and repeated sequences).

The evaluation of these genetic resources will access to the identification of important genome elements which can be mobilised through crossing and transgenic technologies for potentially important allele constitution.

Other important goal of the project is the construction of a mapped marker database in olive using segregating progenies and applying the polymorphisms revealed by different molecular marker systems (RFLP and RAPD). Molecular markers will be linked, in others appropriate segregating Fl progenies of specific crosses, with important phenotypic traits.

This map will serve as the fundamental tool for the manipulation of olive genome to determine a significant advance of the crop products quality, oil and table olives.

Molecular marker technology provides a powerful tool for the measure of genetic potential, genetic relationships and gene chromosomal location. The results of this project will be of great value for the European scientific community, for olive breeders and biotechnologists.
